Antigen Used for Immunization
Morphine–cationized BSA (Mor-cBSA).
Method of Immunization
BALB/c mice immunized with 50 μg of Mor-cBSA in complete Freund's adjuvant intraperitoneally. Booster injections were carried out with incomplete Freund's adjuvant biweekly with 25 μg of immunogen for 3 months.
Parental Cell Line Used for Fusion
Sp2/0-Ag14–mouse myeloma cells.
Selection and Cloning Procedure
Hybridomas were cultured in HAT medium and subcloned several times by limiting dilution.
Heavy and Light Chains of Immunoglobulin
IgG2b (λ light chain).
Specificity
Selected clone toward morphine-BSA was not reactive with BSA. The cross reactivity of anti-morphine MAb with codeine and apomorphine was 100% and 16.5%, respectively. This MAb did not cross react with heroine, papaverine, naloxone, and naltrexone.
