Antigen Used for Immunization
Three equally pooled Dickeya sp. isolated in Hawaii from pineapple stock originated locally and from Costa Rica and Honduras.
Method of Immunization
The bacteria were grown on peptone-yeast agar plates, suspended, and washed two times in PBS, adjusted to OD 0.1 at 600 nm in PBS buffer before use. Mice were immunized intraperitoneally with 0.3 mL of antigen twice a week for 3 weeks before a 2-week rest period, then boosted once more. The fusion was performed 5 days later.
Parental Cell Line Used for Fusion
P3/NSI/1-AG4-1 (NS-1).
Selection and Cloning Procedure
Hybridomas cultured in HAT medium were subcloned thrice by limiting dilution.
Heavy and Light Chains of Immunoglobulin
MAb 2D11 is an IgG1 k light chain. MAb 2A7 is an IgG3 k light chain.
Specificity
MAb 2D11 reacts to most Dickeya species and some other strains of related genera. MAb 2A7 reacts to a genetic subset with a seasonally virulent phenotype of Dickeya sp. isolated from diseased pineapple.
