A60 antigen.
Method of Immunization
Six-week-old female Balb/c mice were given 50 μg of purified A60 antigen in complete Freund's adjuvant intraperitoneally. The next inoculations were performed with half of the antigen in incomplete Freund's adjuvant biweekly in the same way for 2 months.
Parental Cell Line Used for Fusion
SP2/0-Ag14 mouse myeloma cell.
Selection and Cloning Procedure
Hybridomas cultured in HAT medium were subcloned twice by limiting dilution.
Heavy and Light Chains of Immunoglobulin
IgG2 λ light chain.
Specificity
Selected clone toward A60 antigen of Mycobacterium bovis BCG was not reactive towards A60 antigen of Mycobacterium leprae and Mycobacterium paratuberculosis.
Specific Antigen Identified
This MAb recognizes a single protein component in the A60 antigen of M. bovis BCG, with a molecular weight of 65 kDa.
