Abstract

Antigen Used For Immunization
Human serum albumin (HSA).
Method Of Immunization
A pig was immunized with 12 mg HSA (Sigma-Aldrich, Tokyo, Japan) and Freund's complete adjuvant (Difco, Franklin Lakes, NJ) via intramuscular injection. Two weeks post-immunization, a booster containing 12 mg HSA and incomplete Freund's adjuvant (Difco) was administered.
Parental Cell Line Used for Fusion
Not used.
Selection And Cloning Procedure
The phage display method was used.
Heavy and Light Chains of Immunoglobulin
Heavy chain of PKIT-C10 is IGHV1 subfamily.
Light chain of PKIT-C10 is kappa light chain.
Specificity
Phage enzyme-linked immunosorbent assay (ELISA) of the phages displaying HSA-specific scFv showed the binding specificity to HSA.
Specific Antigen Identified
PKIT-C10 had specific binding to HSA.
