Antigen Used for Immunization
Recombinant annexin A2 (AnxA2) protein of Escherichia coli.
Method of Immunization
Six-week-old female C57BL/6 mice were immunized three times with 0.2 mg of recombinant AnxA2 protein at days 0, 15, and 29.
Parental Cell Line Used for Fusion
Mouse myeloma cell line SP2/0.
Selection and Cloning Procedure
The positive clone was selected and cloned three times using indirect ELISA in which plates were coated with recombinant AnxA2 protein.
Heavy and Light Chains of Immunoglobulin
ELISA assay showed the heavy chain of monoclonal antibody SZ-158 is IgG1a subclass; the light chain is kappa.
Specificity
The monoclonal antibody SZ-158 can recognize 36 kDa recombinant AnxA2 protein and inhibit angiogenesis of HMEC-1 cells in vitro effectively.
Specific Antigen Identified
Indirect ELISA assay showed that SZ-158 strongly reacted with recombinant AnxA2 protein.
