Antigen Used for Immunization
RSV A2 strain F protein (F9 epitope, CKIMTSKTDVSSSVI).
Method of Immunization
On three occasions at 10-day intervals, BALB/c mice were injected intaperitoneally with 50 μg of F9 epitope and Lipoplex (O) complex.
Parental Cell Line Used for Fusion
Mouse SP2/O myeloma cells.
Selection and Cloning Procedure
Hybridoma cells selected by screening supernatants with ELISA were subjected to two rounds of subcloning by a standard limiting dilution protocol.
Heavy and Light Chains of Immunoglobulin
Anti-RSV F protein (F9 epitope) monoclonal antibody isotype was identified as IgG2b.
Specificity
Immunofluorescence staining was performed to evaluate whether the obtained anti-RSV F protein (F9 epitope) MAb could recognize RSV F protein specifically.
Specific Antigen Identified
Anti-RSV F protein (F9 epitope) MAb can recognize only RSV F protein in Vero cells infected with RSV A2 strain and has no reactivity with cellular protein.
