PCV2 Cap protein was expressed in Escherichia coli and was purified by immunoaffinity chromatography as immunogen.
Method of Immunization
For the first immunization, equal volumes of antigen and Freund's complete adjuvant (Sigma-Aldrich, St. Louis, MO) were subcutaneously injected into mice. Three booster immunizations with same dose of antigen plus Freund's incomplete adjuvant were conducted at 2-week intervals.

Selection and Cloning Procedure
The culture supernatants were tested using indirect ELISA coated by recombinant S1 protein.
