The in vivo biocompatibility of dextrin hydrogels obtained by polymerization of dextrin-hydroxyethylmethacrylate (dextrin-HEMA) and dextrin-vinyl acrylate (dextrin-VA) are reported in this work. The histological analysis of subcutaneous implants of these hydrogels, featuring inflammatory and reabsorption events, were carried out over a 16-week period in mice. The dextrin-HEMA hydrogel was quickly and completely degraded and reabsorbed, whereas the dextrin-VA degradation occurred slowly and a thin fibrous capsule surrounded the nondegradable hydrogel. The dextrin-HEMA was degraded after 16 weeks with only mild inflammation and a few detectable foamy macrophages around the implant. These events were followed by complete resorption and no sign of capsule formation or fibrosis associated to the implants. The results indicate that the dextrin hydrogels are biocompatible because no toxicity on the tissues surrounding the implants was found. It may be speculated that a controlled degradation rate of the hydrogels may be obtained by grafting dextrin to HEMA and VA in different proportions.

Polyurethane (PU), one of the most versatile biomedical materials, strongly binds iodine, one of the most effective antiseptics, through the formation of a charge-transfer complex. The PU—Iodine complexes were characterized with UV/Vis study and X-ray photoelectron spectroscopy (XPS) analysis. The new materials evoked potent antimicrobial activity against Gram-negative and Gram-positive bacteria (including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus faecium, and bacterial spores), fungi, and viruses, as well as inhibited surface bacterial colonization and biofilm-formation. Based on the Kirby-Bauer test, the antimicrobial effects occurred through the slow release of iodine. The iodine release rate can be controlled by the preparation conditions of the PU—Iodine complex. Trypan blue exclusion analysis indicated that PU—Iodine has excellent mammalian cell viability. The PU—Iodine complexes have the potential for a wide range of medical, dental, and other related applications.

In this report, poly(L-lactide-co-ε-caprolactone), poly(LLA-co-CL) and poly(L-lactide-co-1,5-dioxepan-2-one), poly(LLA-co-DXO) were evaluated and compared for potential use in bone tissue engineering constructs together with bone marrow stromal cells (BMSC). The copolymers were tailored to reduce the level of harmful tin residuals in the scaffolding. BMSC isolated from Sprague—Dawley rats were seeded onto the scaffolds and cultured in vitro for up to 21 days. Cell spreading and proliferation was analyzed after 72 h by scanning electron microscopy and thiazolyl blue tetrazolium bromide (MTT) conversion assay. Osteogenic differentiation of BMSC was evaluated by real-time PCR after 14 and 21 days of culture. Hydrophilicity was significantly different between poly(LLA-co-CL) and poly(LLA-co-DXO) with the latter being more hydrophilic. After 72 h, both scaffolds supported increased cell proliferation and the mRNA expression of osteocalcin and osteopontin was significantly increased after 21 days. Further investigation of these constructs, with lower levels of tin residuals, are being pursued.
