Report: BladeCell Disposable Cuvette for Improved DLS Productivity
A new report from Avid Nano demonstrates how its BladeCell disposable cuvette improves dynamic light scattering (DLS) sample preparation time by a factor of 8 compared with using traditional quartz cuvettes. The 5-μL BladeCell cuvette is precision molded in black polycarbonate and requires no sample preparation beyond centrifuging to reduce the possibility of dust contamination that can affect DLS measurements. To fill, 5 μL of sample is simply pipetted against the walls of the BladeCell liquid trap, where it is retained by surface tension. The prepared cuvette is placed into the temperature controlled sample holder of the Avid Nano W130i, where it can be analyzed by DLS in seconds. After analysis, the sample is recovered by dipping a pipette tip into the liquid trap and removing the contents. Disposal of the BladeCell eliminates the cleaning steps required with quartz cuvettes.
Technical Note: Detecting and Measuring Sub-Nanometer Particles and Molecules
The new technical note (1107-T4) describes how a very small molecule with a low molecular weight (caffeine, MW 194.2 g/mol) was accurately measured at 1.4% concentration in aqueous solution. The results using Avid Nano's W130i DLS demonstrate that the light scattering signal from the dissolved caffeine is sufficient to produce a good-quality correlogram with an intercept >0.2 and highly reproducible data with low polydispersity. The mean radius was consistently reported at about 0.48 nm and molecular weight estimate around 400 Da indicating concentration-dependent self-association into dimers, a known characteristic of caffeine in aqueous solutions.

Application Note: Automated Production of High-Quality Blood Agar
A new application note demonstrates how combining INTEGRA's MEDIAJET Petri dish filler with the DOSE-IT peristaltic pump provides an optimized automated solution for the production of high-quality blood agar. The note describes an automated protocol that minimizes the time that the blood is exposed to hot agar medium and thus reduces thermally induced lysis of erythrocytes. By preparing the blood agar using an INTEGRA MEDIACLAVE media preparator, the dispensing temperature can be set and maintained at low temperature. By combining the MEDIAJET Petri Dish filler together with a DOSE-IT peristaltic pump and a special tubing set for blood agar, the experimenters are able to add blood to the agar immediately before pouring the plate thereby reducing blood cell lysis significantly.

Technical Article: Impact of Product Resistance on Primary Freeze Drying
Authored by Dr. Margit Gieseler of GILYOS and Dr. Henning Gieseler of the Freeze Drying Focus Group at the University of Erlangen, this new technical article discusses how product resistance (Rp) has been recently suggested as a critical product parameter (CPtP) in freeze drying because it instantaneously draws a picture of the (already dried) inner cake morphology at the point of measurement. According to SP Scientific, from a physical point of view, Rp of the dried product layer impacts the resistance to water vapor flow and thus indirectly determines the maximally allowable shelf temperature and primary drying time. In addition to providing an introduction to Rp behavior, the article summarizes the results of trials investigating the Rp of sucrose based on different experimental conditions and using the Auto-MTM feature of the SP Scientific SMART Freeze Dryer software. The authors conclude resistance of the product to vapor flow during primary drying is a CPtP that impacts several process parameters. Lack of information about product performance during primary drying can lead to suboptimal product quality and/or problems during scale-up. The use of MTM allows an assessment of Rp data.

Application Note: The Use of Acumen eX3 for the Identification of the Roles of Protein Kinase C α & δ Isoforms in PMA-Induced Ectodomain Shedding
A new application note from TTP LabTech illustrates how the company's Acumen eX3 laser scanning imaging cytometer can be used to elucidate novel cellular pathways involved in the shedding of the epidermal growth factor receptor (EGFR). The note describes how the high-throughput imaging technology of the Acumen eX3 quantifies and monitors protein expression levels.
According to TTP LabTech, online image capture and analysis, and multiplexing capabilities, allows the Acumen eX3 platform to provide consistent and accurate data from complex biological systems. The shedding of HB-EGF is classically controlled by a disintegrin and metalloproteinase, ADAM17. The imaging technology of Acumen eX3 is used to determine the contributions of additional enzymatic pathways in this process. HT1080 cells, transfected with a modified, alkaline phosphatase-linked, heparin binding EGFR (AP-HB-EGF) are examined by both the traditional enzymatic assay and immunofluorescence for receptor shedding in the presence and absence of inhibitors of ADAM17 and protein kinase C (PKC) isoforms. Experimental data suggest that important contributions are made by the PKC isoforms α & δ. Disruption of EGFR homeostasis is associated with a number of important cancer targets.

Application Note: Mosquito Crystal for Identifying the Structure of Intracellular Glutathione Transport Protein
Entitled “mosquito Crystal for the Rapid Screening and Optimization of Crystallization Conditions of the Bacterial Periplasmic Binding Protein, HbpA and the Elucidation of its Role as a Glutathione Import Protein,” this new application note describes how the mosquito Crystal was used to dispense 100-nL crystallization droplets (50-nL protein sample and 50-nL crystallization condition). This allowed rapid and efficient screening of almost 2000 crystallization conditions, ultimately resulting in the successful crystallization and identification of a novel GSH import protein.

ec23 Stable Synthetic Retinoid
According to AMSBIO, its new ec23 Stable Synthetic Retinoid has been proven to potently induce and reliably control stem-cell differentiation. ec23 enables the robust and reproducible differentiation of stem cells and progenitor cells. With higher stability in tissue culture medium and more potency as a differentiation factor than retinoic acid, ec23 produces downregulation of markers associated with the pluripotent stem-cell phenotype and increased expression of differentiation markers. The differentiation of neural tissues is clearly observed and additionally ec23 has been noted to trigger digit duplication during targeted release in the chick embryo, underlining its potency as a regulator of tissue development. As a potent inducer of neurogenesis, ec23 can maximize the consistency and efficiency of neural stem-cell differentiation and opens up a wealth of applications in basic research screening, toxicological testing, and disease modeling.
The AMSBIO Heparin/GAG Binding Plate and Arrays
According to AMSBIO, products for investigating the protein-binding properties of heparin, heparan sulfate (HS), and other glycosaminoglycans (GAGS) have traditionally been constrained by the need for chemical modification before attachment to inert or derivatized surfaces. The new AMSBIO Heparin/GAG Binding Plate offers a solution to this problem. The specially prepared plate surface adsorbs GAGS without modification while maintaining their protein-binding characteristics. Applications include elucidation of optimum GAG sequences and sulfation patterns for binding to proteins of interest, analyses of sulfotransferases and endosulfatases, and detection of inhibitors or enhancers of protein-GAG recognition. The new plate also can be used for studying receptor binding to growth factors or chemokines bound to surface-immobilized GAGS. The ELISA-type format of the Heparin/GAG Binding Plate uses protein reagents economically and is compatible with automated laboratory systems.

ProteOn HTG Sensor Chip and Reagent Kit
For use with the Bio-Rad ProteOn XPR36 system, the new ProteOn HTG Sensor Chip's surface chemistry has increased binding stability and specificity for His-tagged proteins compared with chips from other surface plasmon resonance (SPR) system providers. Additionally, the HTG Sensor Chip's ability to regenerate and reuse HTG chips multiple times can reduce the cost of generating data. The ProteOn HTG Sensor Chip's Tris-NTA complex contains three NTA molecules for improved binding stability and selectivity to His-tagged molecules. This results in better data quality and reduced ligand decay. Furthermore, binding via polyhistidine tag is reversible, so the ligand can be removed from the surface and the chip can be reused 10 times or more. This increases reproducibility by eliminating chip-to-chip variability, and it decreases cost per data point.
To optimize the polyhistidine-tagged protein interaction workflow, Bio-Rad has also introduced the ProteOn HTG Reagent Kit for use with the ProteOn HTG Sensor Chip. The kit includes NiSO4 for chip activation and an EDTA solution for chip regeneration.

The CytoPatch Instrument is the first automated patch-clamp platform in which the patch-clamp process takes place inside a three-dimensional (3D) quartz glass microchip with integrated microfluidic channels. The new Versa 3D's configurable platform provides high-resolution, 3D imaging and analysis on a wide range of sample types. Versa 3D is available with either high vacuum only or high- and low-vacuum electron imaging hardware. According to FEI, this new member of the Titan G2 series of S/TEM (scanning/transmission electron microscopes) can deliver atomic resolution elemental maps in minutes and adds new capabilities for applications in materials science, chemistry, and nanotechnology. This new microscope includes features of the Titan G2 series, including the X-FEG high brightness gun and the next-generation DCOR probe corrector, achieving spatial resolution of 0.8 Å in STEM and 0.9 Å in TEM, while still maintaining a large, more flexible working space in the sample area, enabling applications requiring high-specimen tilt angles. For example, this large tilt range, when combined with ChemiSTEM Technology's symetrically distributed 4-SDD detector architecture permits EDX Tomography. Thus, 3D elemental composition can be obtained with standard FIB-prepared lamella samples mounted in standard holders in an S/TEM.

This new system offers image deconvolution/deblurring for ultra-sharp images and provides new fluorescence imaging capabilities. Virtual slide creation can be achieved by setting the slide in place and pressing Start. According to Olympus, the system creates outstanding bright field slide scans up to three times faster. The VS120 can scan up to 100 slides at a time at very high fidelity, so that users anywhere can view and fully navigate high-resolution images of entire microscope slides or slide areas using a computer. It comes equipped with a 5-megapixel CCD bright field camera that improves bright field scanning speeds while delivering superb color reproduction and ultra-high resolution.
Acquiring sharp images at multiple depths using six or more fluorescence labels is possible, thanks to Olympus Fluorescence Virtual-Z capability. The software adjusts for the potentially different focal positions of various wavelengths in multichannel acquisition. Automated batch scanning can be accommodated with fluorescence samples and individual shading correction is available for each color channel. For chemical synthesis on the mg to kg scale, this system incorporates a wide range of temperatures (−15 to +250 °C), pressures, and flow rates, and can be used with solid- or solution-phase chemistry. According to Syrris, its high chemical compatibility enables a broad range of reactions. The Asia 220 can carry out a single reaction from start to finish, providing “walk-away” automation, without the need for a PC.
The Asia 220 enables automated synthesis. Users enter the reactor volume, desired temperature and pressure, collection volume and desired ratio of the reagents, then press start. The system automatically sets the flow rates, injects the correct volume of reagents, controls reaction temperature and pressure, diverts the waste, and starts and stops the collection when the reaction product reaches the Product Collector.

CompacT SC Automated T-Flask Cell-Culture System
According to TAP, its new CompacT SC can be used to aseptically culture multiple different stem cell lines in parallel using T75 and T175 flasks, has a number of features that make controlling the growth and differentiation of stem cells easy. The system includes automated cell imaging and analysis that allows the CompacT SC to be programmed to monitor cell morphology and growth curves. It also permits the system to bet set up to respond to any preprogrammed changes by automatically passaging cells and adding new media, reagents, or vectors at appropriate times. This means it can maintain cell lines in the state needed, either as pluripotent cells or during cell differentiation and transfection. Processes can be performed at anytime of the day or night, without users having to be in the laboratory.
The CompacT SC also comes with a Low Volume Reagent Dispensing Option and an Incubator Oxygen Control Option to enable optimal conditions for stem cells by automatically setting the system to pipette chilled growth factors or transfection reagents at low volumes, and provide reduced oxygen levels to mimic in tissue conditions.

Gas Addition Module for FlowSyn
According to Uniqsis, this new module enables fast, controllable presaturation of liquid reagents with a wide range of gases and so promotes efficient gas-dependent reactions in flow, such as hydrogenation, ozonolysis, carbonylation, and direct synthesis of carboxylic acids. The pressurized tube-in-tube design of the Gas Addition Module overcomes the problem of undissolved gas bubbles by ensuring continuous interaction between the gas and liquid at every point along its length. The tube-in-tube design is based on semipermeable membrane technology, whereby the semipermeable inner tube containing the liquid (typically a solvent) is bathed by a stream of pressurized gas that is enclosed within a thick-walled impermeable outer tube. The pressurized gas is able to cross the semipermeable membrane of the inner tube and dissolve into the liquid carried within. However, because of the semipermeable nature of the inner tube material, the liquid is unable to cross in the opposite direction. The Gas Addition Module is compatible with the Uniqsis FlowSyn and other continuous flow reactors. It can also be used as a reactor in its own right.

Maximizing fume cupboard coolant use, the DrySyn Water Manifold converts a single water tap into three condensing streams, and collects the outlets into one drain tube. With the main body constructed from anodized aluminum, the manifold is designed to be clamped to fume hood scaffolding in either vertical or horizontal alignment. Quick release plastic connectors enable rapid reaction setup and dismantling, with sealing on both the tubing and the manifold side. The medium capacity Allegra X-30 Series Benchtop Centrifuge from Beckman Coulter is a compact, 18-in.-wide instrument that is available individually or packaged with the appropriate rotors and labware for a range of applications. The new Falcon MEGA SNMP/HTTP agent enables remote control, monitoring, and operations system shutdown solutions. The new agent consists of stand alone external or an internal circuit card that fits into the communications option slot of any Falcon SG, SSG, or FN Series rackmount and tower uninterruptible power supply (UPS). The agent allows for the connection to any Ethernet LAN. Two USB ports on the agent support the connection of an optional 802.11G Wi-Fi dongle providing wireless LAN connectivity. Any number of RIC20s can be used via the RS232 I/O port on each unit and PC serial ports. Complex temperature/time profiles created by users can be run by leaving the computer attached to the RIC20. Any number of units can be set to any single temperature via a single computer serial port. The unit(s) will continue to run the temperature instructed until told otherwise. The computer can then be used to instruct the next RIC20.

STR Profiling Service for Human Cell Line Authentication
To verify the identity of human cell lines via short tandem repeat profiling (STR profiling) cells are applied to an FTA card provided with an FTA Sample Collection Kit from American Type Culture Collection (ATCC), which eliminates the need to ship samples on dry ice. ATCC will amplify the DNA using the Promega PowerPlex 16 HS System and analyze the results, which deliver via e-mail within 5 days from receipt of samples.
ATCC offers two levels of service: Basic and Verified. The Basic STR Profiling Service (135-X) includes a signed Cell Line Authentication Report containing an STR allele table and electropherograms that support the allele calls at each locus. The Verified STR Profiling Service (135-XV) includes everything in the Basic service, plus known reference profiling against the ATCC STR Profile database, and a comprehensive interpretation of the results.

Using Agilent SureSelect Target Enrichment technology with the Beckman Coulter SPRIworks Fragment Library System for automated library construction and the Biomek Laboratory Automation Workstation for liquid handling provides high-quality, reproducible samples ready for sequencing. Sequencing is performed on the Illumina GAII and HiSeq 2000 platforms. LIMS sample tracking also enables large numbers of samples to be processed at the same time.

Micro Determination of Fluorine
Operating in an ISO 17025 and GMP accredited environment, Warwick Analytical Service (WAS) now offers proprietary analytical procedures to enable accurate and precise data to be produced for all types of samples including organic samples with trace levels of Fluorine. From extensive method validation, typical results are within the ±0.3% absolute limits considered acceptable for micro chemical analysis. The WAS method can detect 0.01 ppm fluoride, which is equivalent to 0.02% in a 10-mg sample. WAS has developed special procedures to remove interferences from samples containing boron, calcium, or sulfur.
Analysis of Fluorine is carried out by combustion of an accurately weighed sample in a silica oxygen flask over an accurately measured volume of water and TISAB buffer solution. A fluoride ion selective electrode is used to directly measure the potential of a range of fluoride solutions. The potential of the combusted samples are measured under the same conditions. The concentrations of the sample solutions are calculated from the derived calibration curve. Sets of work typically include certified reference materials or organic analytical standards. Results from these standards and the electrode response are monitored to detect trends in the method.
