Antigen Used for Immunization
1-(heptyl-7-carboxylate)-3-(1-naphtoyl) indole-KLH conjugate.
Method of Immunization
One hundred μL of prepared adjuvant emulsion (1:11-(heptyl-7-carboxylate)-3-(1-naphtoyl) indole-KLH conjugate (100 μg per mouse) and complete Freund's adjuvant) were injected intraperitoneally into female A/J mice (8 weeks of age). The mice received the same injections 2 weeks later. Following another 2-week interval, the mice were injected intraperitoneally with 100 μL booster solution containing 100 μg 1-(heptyl-7-carboxylate)-3-(1-naphtoyl) indole-KLH conjugate.
Parental Cell Line Used for Fusion
Mouse myeloma cell line P3X63-Ag8.653.
Selection and Cloning Procedure
Selected by HAT media, positive clone identification by ELISA.
Heavy and Light Chains of Immunoglobulin
Heavy chain, g1; light chain, k.
Specificity
ELISA indicated that MAb NT2 recognized 3-(1-naphthoyl) indole derivatives.
Specific Antigen Identified
3-(1-naphthoyl) indole derivatives such as 1-methyl-3-(1-naphthoyl) indole, 1-ethyl-3-(1-naphthoyl) indole, and 1-octyl-3-(1-naphthoyl) indole.
